The way I know it, at the very beginning of the first night, Cupid (or Amor or Yumi) goes around the table and pat two people on their shoulders. After Cupid is "asleep" again, the Game Master "The lovers wake up, recognize each other and go back to sleep." (Of course they do as said). After that, the game continues as normal (since we normally play with cards to select the rules, the hunter etc. have to do the same for the GM to know the roles).
Actually it gets really interesting if the lovers are in different parties, for example the detective and one of the Mafia, because the game is going to get more and more twisted, and I wouldn't suggest to change that possibility...

And I'd suggest another role of the Mafia (since we have two people):

The Slanderer (lit. translation of German: Rufmörder). His role is simple - he chooses one player at night (after the killing he stays awake, the other Mafia do not know his decision). Said player is done no harm at all - but if the detective investigates him, he'll get the opposite result he'd normally get(!). This, of course, limits the power of the detective, since he can't be completely sure his result is right. Again, the game gets more twisted.
I'm sorry, I can't think of any DC character to fit him. Perhaps Vermouth, the master of disguise?

Oh, and I wouldn't want 1412 to be the Trickster. After all, the Trickster gets nothing done and is a burden to everyone - that's Eisuke (and, BTW, I don't like him as well - and that's why I suggested him)!
And, as mangaluva said, Agase would make a good doctor. After all, he helps the detective (Conan) to defeat the bad guys (through his inventions  ;D). Not to mention that Araide may be Akai (Hunter) or even one of the BO (Mafia) as far as I know...

The thing about Cupid is that if I was to make Cupid choose, either we'd need to wait for Cupid to choose before starting the game or we'd have to hope Cupid is on immediately as the game starts. It's one of those things that works better in person, but on a message board where we're all in different time zones, I can't exactly expect the random Cupid to be on right as the game starts. Although, I could leave in the possibility of people being both the lovers + a different role anyway.

You make a good point about the trickster though, so maybe he'll be Eisuke after all. :P

While Agasa is in fact a Doctor, I'm not exactly sure I'd want him saving my life. He's an inventor, not really a medical doctor. As for Araide's identity, it's not really a huge deal, because if we do the thing with lovers possibly being double-roled, we could get equally strange things like Akai x Shinichi loving going on.

Xcommando wrote: how bout cupid choses at night so we dont have to wait
that would be doable, but would result in the lovers not knowing each other at the start. So, I'll handle it like this:
-Cupid chooses during the night
-Lovers learn of each other at the dawn of the day
-If one of them is dead at this point, the other one will kill themselves anyway
So, things might get unfortunate if a Mafia member kills someone the first night, who cupid designated as his lover. But this is also sorta interesting, so I'll do it if you're all fine with it.

Also, as of the current moment, these are the roles (the PMs each person will receive at the start of the game, telling you your role)

So, if you have any problems with this list, speak up because we got 4 hours to go till I'm aimin to start this! :O!
Spoiler:
You are Shinichi, the Great Detective!
(Detective)
With your deductive abilities, you are able to investigate one person a night, and find out whether or not they're part of the Black Organization. They're lurking somewhere in this town, and you're the only one who knows how to look for them!

Watch out though, you might not be able to get any investigating done if that punk Eisuke the Trickster interferes with your investigating. What's his deal anyway, huh?

You are Gin of the Black Organization!
(Mafia, Slanderer)
You have come to Beika to hunt down a target that's eluded you for quite some time. Unfortunately, Vodka lost the orders somewhere. What imcompetence! The only solution to this problem? You have to kill everyone in the town, to make sure you find whoever this person is, and put an end to them for good.

As Mafia, every night you are able to choose one person to kill. Discuss who to kill with Vodka, he's actually pretty good at killing people.

As a Slanderer, you're able to use your gift of intelligence to manipulate information. Every night, along with killing someone with Vodka, you are able to choose another person to slander (this is optional), causing anyone who investigates that person that night to think that they're a member of the Black Organization. This can also be used on a member of the Black Organization to make the detective think that they're good. What would the Black Organization ever do without you?

As Mafia, every night you are able to choose one person to kill. Discuss who to kill with Gin, he's actually pretty smart.

You are Araide, Doctor Extraordinaire!
(Doctor)
Well, you're a doctor anyway. Most people don't actually know you, but that's how you like it. As a doctor, every night you can creep into one person's home, and perform medical miracles on their body. If this person was a victim of an attack that night, you will know. However, they will not, as your medical prowess leaves their body free of any marks or injuries. And then, quietly as you entered, you leave- and nobody knows you were there.

You are Shuichi Akai, the Badass!
(Hunter)
Some people say you can get shot in the head and not die. Well, that's not really true. You're not quite death-proof, but you're pretty resilient, so you don't die immediately. And ever since Akemi died, well, you're pretty spiteful too.

As the Hunter, if you get lynched, you manage to kill the person *you* wanted to lynch right before you die. That's pretty awesome, but everyone knows you're awesome anyway.

You are Yumi, the Matchmaker!
(Cupid)
You were the one who got the lovers together. Because of this, you know who they are. You also know that they have this weird idea about being alone in a complete lovers' paradise, and you wonder how far they'll go to make sure that in the end, they're alone...

Otherwise, you're basically a normal citizen of Beika.

(You know the identity of the Lovers)

(While you're technically a cop, you're just a traffic cop and you're not competent enough to investigate the Black Organization, leave that to Shinichi)

You are a Lover!
(Lover)
You live for your significant other, so much that you don't know what you'd do without them. So much, that if they died, you'd kill yourself due to grief.

On the other hand, if you and your significant other are the only two alive at the end, you both win! Huh, what about Yumi, the matchmaker? Well, if she's alive, you and your significant other can't be alone in your perfect lovers paradise now, can you? Yumi must die.

(You know the identity of your significant other and Yumi)

You are Eisuke, the Trickster!
(Trickster)
Nobody likes you. Not a single person. But you don't really know why. So, to spite them, you make sure that nobody can get anything they want to do done, by secretly interfering with the things they do.

As the trickster, every night you can choose one person to trick. If this person ends up being the Detective, he won't get his report back. If it's either member of the Black Organization, they won't be able to kill anyone. If it's the Slanderer, he won't be able to slander anyone, either. If it's the doctor, well... let's just say that injecting people with water doesn't actually do anything useful.

You are a citizen of Beika.
(Townsperson)
Well, look at you. You're an average person. At least, compared to the rest of the characters. You probably have some interesting backstory, but we haven't gotten to that episode yet, so poor you.

Don't let that stop you from pretending to be someone else during the day, though.

Concerning the Slanderer:

He might indeed slander anyone, but the slanderer can also "slander" any Mafia member, therefore making them "good" towards the detective. Just an important side-note (therefore, even if the Detective gets good, it mustn't mean good. With your method, it still would.).
If  ;), :D, ;D, ::), :P, :-X, :o or >:D are attached, that paragraph may not be 100% serious. Seriously.

touché, I'll add that into the things.

Oh, and another thing:

You can also give the commoners names from DC characters as well. It'll just sound boring if we get the pairing Shinichi x Commoner_1. But if they have names from DC characters, we might get Shinichi x Ran (or Haibara or whoever), and things will get more funny. And of course, if Ran (or Ai or whoever) is killed, we can mourn for her/him as well.

And you might illustrate the murders as well, so not only "mangaluva alias Yumi is dead" but "Yumi, also known as mangaluva, has been found dead in heir apartment with the notes of the Ode To Joy impaled by a knife through her heart. The people of Beika have gathered to determine who committed this heinous act."
Yes, this is the task of the Game Master (although you might also give this work to the Mafia and let them specify the circumstances of the death).

True about the Commoners, I'll give em actual names when the game starts.

I was gonna illustrate the murders, of course. Playing mafia is only half the actual game, the other half is in the descriptions of the situations and the accusations etc. Mafia people can feel free to say how they kill their victim as well.

Some possible commoners you could be:
Spoiler:
You are Ran.
(Townsperson)
You're Ran, lucky you! What does that mean for you? Well, not a whole lot, because even though you're one of the main characters you don't have much of an active role in the story. You can cook a champ, though. You're also good at karate, but not when you're asleep. Unfortunately for you, that's when the Black Organization makes their moves.

Don't let that stop you from pretending to be someone important, though.

You are Kogoro.
(Townsperson)
You are Sleeping Kogoro, the greatest detective who ever lived. Or so you think. You're actually horrible at it, and you can't solve anything on your own, so don't bother trying.

Don't let that stop you from pretending to be a great detective (or someone else important), though.

You are Haibara.
(Townsperson)
You're Haibara, the former member of the Black Organization who fled to Beika in the body of a child. Recently though, you've been smelling some pretty bad BO around town... time to put on that baseball cap and some sunglasses and hope they don't recognize you.

But hey, maybe if you pretend to be someone else, they won't realize you're Shiho.
